Heading into the crucial Matchday 33 encounter, Blanket Mine were sitting at the top of the standings with 78 points, while Kwekwe United trailed on 73.
The visitors needed only a draw to secure the championship, while Kwekwe United were desperate for victory to keep alive their hopes of an immediate return to the top flight following relegation last season.
The achievement marks a significant milestone for the Gwanda outfit, which has enjoyed an outstanding campaign under coach Luke Petros Jukulile.
Blanket Mine have lost just once in 33 league matches, demonstrating remarkable consistency throughout the season.
Speaking after the match, Jukulile expressed delight at guiding the club to the Premier Soccer League, describing the achievement as one of the highlights of his coaching career.
Blanket Mine's promotion will bring top-flight football to Gwanda and provide another opportunity for football supporters in Matabeleland South to watch Premier Soccer League action.
